The Ho Lab in the Department of Microbiology & Immunology at Columbia University is seeking a dedicated and enthusiastic Technician B to join our team. We use cryoEM and cryoET to explore how membrane protein complexes mediate host-pathogen interactions in the malaria parasite Plasmodium falciparum. Successful candidates will join a vibrant research community and receive training in the state-of-the-art electron microscopy facilities at Columbia, including:
Titan Krios microscopes with a Gatan BioQuantum energy filter, K3 Summit direct electron detector, and Volta phase plate
An Aquilos cryo-FIB/SEM microscope, Leica cryo-correlative fluorescence light microscope, Zeiss LSM800 cryo-confocal with AiryScan detector
An F20 screening microscope with a K2 Summit direct electron detector
Under the supervision of the Principal Investigator, the Technician B will provide assistance in projects involving cloning and culturing blood-stage P. falciparum parasites for single-particle cryoEM structure determination of native membrane protein complexes purified directly from malaria parasites and cryoelectron tomography of the host-pathogen interface in parasite-infected red blood cells.
Duties will include:
Maintaining and culturing P. falciparum parasites for structural studies
Assisting with creating P. falciparum parasite lines
Assisting with purifying proteins from parasite lysates
Ordering and maintaining laboratory supplies
Performing other related duties necessary to the functioning of the laboratory, research project and/or work unit
B.S. degree in microbiology, biochemistry, molecular biology (or related field) or equivalent in education, training and experience.
A strong background and proven track record in one or more of the following areas is preferred: microbiology, molecular biology, tissue culture, protein biochemistry.
Previous experience in CRISPR-Cas9 gene editing and/or culturing blood-stage P. falciparum parasites is highly desired.
Strong communication, interpersonal and organizational skills, and excellent attention to detail.
