The objective of our research is to understand how the collection of red/far-red light-absorbing photochromic photoreceptors - phytochromes - enable perception of light by plants and microorganisms through analysis of their 3D structures using atomic level methods such as x-ray crystallography and cryo-electron microscopy. The advertised position will aid in these efforts by expressing and purifying recombinant wild-type and mutant versions of these photoreceptors, assist in their biophysical and biochemical analyses, and help develop phytochrome samples for subsequent structural studies.
PRIMARY DUTIES AND RESPONSIBILITIES:
Express recombinant phytochromes in Escherichia coli-based synthesis systems.
Purify recombinant phytochromes using various chromatography approaches, including FPLC.
Define the purity of the phytochrome preparations via UV-visible absorption spectroscopy and SDS-PAGE.
Analyze the photoconversion and thermal reversion kinetics of the phytochrome preparations via spectroscopic assays.
Generate mutant forms of phytochromes by site-directed mutagenic strategies for subsequent purification and analysis as above.
Help with laboratory maintenance and ordering of supplies.
Perform other duties as assigned.
B.S degree in biology, biochemistry, or related areas and 1 year of experience or equivalent education/experience to equal 5 years.
Prior research experience helpful, especially related to microbiology and bacteria growth.
Excellent technical and communication skills helpful.
This position is full-time and works approximately 37.5 hours per week.
